• Multi-parametric analysis can provide a powerful tool for identification and functional interrogation of discrete cellular populations at the single cell level. Rational design of a multi-color application is dependent upon the ability to properly pair the available technology to the scientific question under investigation. Understanding the spectral characteristics and limitations of each of the fluorochromes used is essential to avoid the pitfalls associated with dye interactions resulting in aberrant staining patterns. • Over the last years several new technologies and instruments in the field of high-speed cell sorting have been developed and flow sorters have become a widespread and vital resource in the biological sciences and beyond. This presentation will give an overview on specific features and first experimental experiences working with the new MoFlo XDP. A special focus will be put on high-speed, rare event analysis and sorting. The principles behind ‘particle sorting’, multi parameter setup and some practicalities for a successful sorting experiment will be discussed.
